Application of staining techniques to improve the viability assessment of turbot (Psetta maxima) ova ArchiMer

Staining-dye procedures were tested on the turbot ovule in order to develop a rapid technique for determining ovule viability. Neutral red and Trypan blue dyes provided better assessment of the proportion of viable or dead female gametes when inducing fertilisation.

Cultured heart cells from oyster: an experimental approach for evaluation of the toxicity of the marine pollutant tributyltin ArchiMer

European Community regulations on chemicals promote alternative methods to test substances presenting potential risks for the environment. In the present work, cultured atrial cells isolated from oyster (Crassostrea gigas) were used as an experimental model to investigate the toxicity of tributyltin (TBT) after short-time exposure at concentrations representative of those that can be measured in seawater, marine sediments and/or bivalves bioaccumulating this pollutant. In vitro and in vivo assays produce values of the same order of magnitude for both animal/cell survival and heart/cardiomyocyte beating rate. This work is part of the program, "recruitment determinism in scallops" initiated in 1992 by the "Contrat de Baie". The aim of this research was to explain interannual abundance fluctuations observed for Pecten maximus, in the Bay of Brest. To this end an analytical approach to the life cycle of the species was undertaken to determine its critical phases. A phase may be considered as "critical" if the mortality rate at its level varies from one year to the other. Using data from 1989 to 1994, a precise time-table of the "physiological year" of the scallop in the Bay of Brest was determined. Little information is available on viral infections that affect bivalve molluscs. Such a lack of data is due ta a certain inadequacy of the diagnosis methods that are employed when massive mortality events occur. Most laboratories involved in mollusc pathology still analyse samples through light microscopy. World-wide, there is thus currently a lack of information concerning the occurrence of bivalve herpesvituses. This is probably due to the lack of suitable diagnostic tools. The basic method for identification and examination of suspect samples is predominantly histopathology. La coquille Saint-Jacques est un bivalve largement exploité sur les côtes françaises. En Rade de Brest, cette espèce présente un intérêt commercial tout particulier du fait de présenter une noix (muscle adducteur) et un corail (organe reproducteur) bien développés toute l'année. Elle fait l'objet d'une pêcherie spécifique et très contrôlée. Durant la phase préparatoire du contrat de Baie de la Rade de Brest, des travaux financés par le programme Rade ont été menés à l'écloserie du Tinduff dans le but d'améliorer le soutien au stock naturel exploité par cette pêcherie. The quality of the scallop embryos is determined by the spawners characteristics as well as by the external factors they support during the maturation and the spawning phases. All these parameters are not still well controlled during the artificial conditioning in the hatcheries. As a consequence, the quality of the embryos is drastically variable and with them, the larvae performances in terms of survival and growth rates. It would be interesting to get a simple and quick test which would allow the estimation of the spawns quality. The embryos density, evaluated by isopycnic centrifugation of Percoll medium, is well adapted to this objective. L'environnement gonadique des cellules sexuelles, le déroulement de la gamétogenèse, le devenir des gamètes formés et leur qualité au moment de l'émission ont été étudiés chez Pecten maximus sur le plan ultrastructural et histochimique. Sperm features in turbot (Scophthalmus maximus): a comparison with other freshwater and marine fish species ArchiMer

This paper presents some features of turbot sperm and compares them with data reported in the literature on other freshwater and marine fish. In captivity, turbot spermiation period lasts 6 months. When compared to other fish species, turbot males are characterized by low values of maximum gonadosomatic index (0.6-0.8%) and are poor sperm producers in terms of volume (0.2-2.2 ml), concentration (0.7-11.0 x 10(9) spermatozoa/ml) and total sperm number released per stripping (0.2-12.0 x 10(9) spermatozoa). Compared to most freshwater species, the duration of cell movement is long (1-17 min). The spermatozoon of turbot can be considered to be of a primitive type.
